摘要:
The combination of an aging population and improved survival post myocardial infarction have contributed to a rapid rise in the incidence of heart failure worldwide.  Yes, despite the associated morbidity and mortality as well as economic burden of heart failure, few therapeutic options exist to aid failing heart muscle.  For the past decade, we have focused on understanding the organization and activity of ventricular cardiomyocytes.  Our approach is to learn trafficking pathways of membrane ion channels, the membrane microdomains that support channel delivery, and the cytoskeleton based trafficking delivery routes.  Highlights are we have defined Targeted Delivery (Cell 2007, PLoS Biology 2010), by which ion channels travel to specific membrane microdomains, the organization of membrane structures which accept channel delivery (Nature Medicine 2014), the formation of de novo proteins to aid in trafficking (Cell Reports 2013), and alterations in disease (JCI 2010, Circ Res 2012, Heart Rhythm 2012, Traffic 2014).  This lecture will focus on cardiomyocyte organization and the role of key proteins such as products of alternatively translated GJA1 and cardiac cBIN1 that are responsible for the intracellular trafficking that continuously occurs in each heart cell.
